Glioblastoma is a highly aggressive form of brain tumor. MGMT gene promoter methylation is observed in about 50% of glioblastoma patients.
MGMT is a gene that encodes O6-methylguanine-DNA-methyltransferase enzyme involved in DNA repair. The active form of MGMT provides repair of damaged DNA caused as a result of the action of alkylating agents. Upon loss of MGMT function due to epigenetic methylation of the promoter, cells are left with a reduced ability to repair these damages, leading to their cell death. This makes the action of alkylating chemotherapy drugs more effective.
Determination of MGMT promoter methylation status in patients with glioblastoma is used as a biomarker with prognostic and predictive value and according to current recommendations is a necessary routine test for treatment selection in adult patients. Patients with methylated MGMT have better overall survival rate and respond better to chemotherapy with alkylating agents (temozolomide).
EpiDirect® MGMT qPCR Kit offers an innovative approach for direct analysis of MGMT promoter methylation status without the need for bisulfite conversion of DNA.
As part of the kit features are:
- Quantification of MGMT methylation status expressed as % methylation frequency
- Ready-to-use format - just add DNA to a ready-prepared master mix pre-aliqoted in PCR tubes
- Two detection channels - FAM and HEX, which makes it compatible with most Real-Time PCR machines
- Quick analysis - 1 hour and 30 minutes.
Analysis of MGMT methylation status is recommended to be carried out before starting treatment - when choosing the type of therapy - radiotherapy or chemotherapy, as well as when choosing the specific type of chemotherapy agents.
