The PCR method is the gold standard in detecting and proving infection caused by Bordetella pertussis, distinguished by the greatest sensitivity and speed of execution. The PCR method allows proof of active infection and carriage without pronounced clinical symptoms, characteristic of vaccinated individuals.
Serological methods are another testing option that allows the establishment of the immunological status and the presence of an active infection, but at a later stage of its development, which is why they carry the risk of false-negative results when the infection is in the initial stage.
Several factors affect the analytical characteristics of the assays and the final results obtained from them, one of the most critical and determining factors being the method of sampling.
In this regard, we are sending information with up-to-date recommendations and guidelines for sampling:
Sampling for PCR analysis:
- It is recommended that samples for analysis be taken within 2-3 weeks of the onset of clinical symptoms and preferably before the initiation of antibiotic therapy
- Materials for PCR diagnosis of pertussis are nasopharyngeal secretions taken from the ciliated epithelium of the upper respiratory tract from suspects and contacts of pertussis infection
- Clinical specimens are collected using a dry synthetic sterile swab without transport media. Cotton swabs are not allowed to be used due to the possibility of inhibiting the PCR assay
- Immediately before DNA extraction, the swab is resuspended in sterile saline or molecular biology water (DNA, RNA, DNase, RNase free) and subjected to the extraction process
For your analysis, we recommend:
- Nasopharyngeal Flocked Nylon Fiber Swabs - https://www.ridacom.com/en/products/view/7485
- VIASURE Bordetella Real-Time PCR Detection Kit - PCR kit for simultaneous detection and differentiation of B. pertussis, B. parapertussis, B. holmesii species - https://www.ridacom.com/en/products/view/7697
Sampling for serological analysis:
- Samples for analysis can be collected between 4 and 12 weeks from the onset of clinical symptoms, with the optimal sampling period being between 4 and 8 weeks when antibody titers are assumed to be at their highest.
For your analysis, we recommend:
- SERION ELISA classic Bordetella pertussis – IgA, IgG & IgM
Quantitative and qualitative immunoassays for the detection of human antibodies in serum or plasma directed against Bordetella pertussis for laboratory confirmation of whooping cough. Differential detection of distinct antibody classes supports the categorization of disease stages. IgM antibodies for the diagnosis of acute primary infections, especially in children. Detection of IgA and IgG antibodies for laboratory confirmation of whooping cough, as well as for differential diagnosis in atypical pneumonia. IgA and IgG antibody activity results are expressed in IU/ml against the first WHO international standard - https://www.serion-diagnostics.de/en/products/serion-elisa-classic-antigen/bordetella-pertussis/
- SERION ELISA classic Bordetella pertussis Toxin – IgA & IgG
Provide specific and differential diagnosis of Bordetella pertussis. Whales use Pertussis Toxins (PT) as an immunogen to distinguish an infection caused by Bordetella pertussis from that caused by other Bordetella species - https://www.serion-diagnostics.de/en/products/serion-elisa-classic-antigen/bordetella-pertussis-toxin/
